    Subtelomeric secondary metabolite (SM) gene clusters are frequently surrounded by DNA
    repeats and transposon-like elements. The A. nidulans penicillin cluster, 30 kb from the telomere of
    chromosome VI, is bordered by such elements. Deletions of penicillin telomere proximal and distal
    border regions resulted in decreased penicillin production. A 3.7 kb distal region called PbIa,
    consisting of the putative transposable element DNA-2, was examined further where its replacement
    by a pyrG marker presented a similar phenotype as loss of the global SM regulator LaeA, resulting in a
    decrease in penicillin gene expression and product formation. In contrast, placement of the pyrG
    marker on either side of PbIa had no effect on penicillin synthesis. A requirement for PbIa in penicillin
    production was also apparent in a histone deacetylase mutant, ?hdaA, enhanced for penicillin
    production. Trans complementation of the PbIa element near and within the terrequinone A cluster on
    chromosome V did not restore penicillin biosynthesis or increase production of terrequinone A. Taken
    together, this data provides evidence for transposon involvement in SM cluster regulation.Œ0??

    Penicillin Production by immobilized cells of Penicillium chrysogenum NRRL 824
    The production of penicillin by free and immobilized cells of Penicillium chrysogenum was investigated. The cells were immobilized using, synthetic sponge, cotton balls, loofa pieces as well as calcium alginate. The maximum amount of penicillin reached up 128±10.37 µg/ml was obtained, using loofa sponge as a carrier as compared to free cells production (19.4±2.05 µg/ml). In addition to 31.9±4.5 µg/ml and 48.9±7.2 µg/ml of penicillin obtained by using the cells immobilized by sponge cubes and cotton balls respectively. However, the use of calcium alginate beads produced lower yield (6±0.53 µg/ml) than obtained by free cells but different numbers of used loofa pieces, sponge cubes and cotton balls greatly influenced the production of penicillin. The highest antibiotic yield was obtained by using 16 equally sized sponge cubes, 20 cotton balls and 5 loofa pieces (31.9±4.5, 48.9±7.2, and 136.9±10.37 µg/ml respectively).
    During repeated batch fermentation process, loofa-immobilized cells exhibited higher penicillin production (340 µg/ml) at the 4th fermentation cycle than other immobilized cells under similar experimental conditions. Therefore, immobilization of P. chrysogenum cells using loofa can be successfully used to increase penicillin production more than other carriers.
